Neurogliaform development
To study NGC embryonic development, we collected several single-cell transcriptomic datasets covering the Preoptic Area at E14.5 (flash tagged progenitors, genetically fate-mapped NGCs and WT cell landscape).
Using NGC machine learning model trained on postnatal data, we identified embryonic NGC progenitor and postmitotic cells. Being able to rule out type and subtype lineages, we discovered a small subset of genes that are conserved throughout NGC development, including the transcription factor Tox2. Interestingly, NGC subtypes express Eph receptors in a combinatorial manner (Epha3 vs Epha4).
